About our HMWoTau ELISA

Our highly sensitive novel ELISA detects HMWoTau species in a disease specific manner. Our ELISA (APRI.HMWoTauELISA) employs APNmAb005 mouse monoclonal antibody that recognizes HMWoTau (about 30 nm diameter), labelled by Thioflavin (Fig. 1). APRI.HMWoTauELISA detected Alzheimer’s disease (AD)-derived HMWoTau species, in a dilution dependent manner (Fig.2A) and at Braak Stage V and VI of AD tau pathology in the frontal neocortex of AD brain (phosphate buffer saline extractable fraction) (Fig. 2B). APRI.HMWoTauELISA standard curve using recombinant human tau-derived HMWoTau showed quite a good linearity with Lowest Limit of Quantitation 0.3 pg/well (Fig. 3) (all figures are adopted from FASEB J. 2024 Nov 30;38(22): e70160). APRI.HMWoTauELISA also detects the HMWoTau species derived from transgenic mice bearing human Tau and from cell cultures.
